Purpose
The Vitamin B12 and Folate Test measures the levels of vitamin B12 and folate (vitamin B9) in the blood. These essential nutrients are critical for red blood cell formation, DNA synthesis, and proper nerve function.
Low levels of either vitamin can lead to megaloblastic anemia, neurological complications, and cognitive impairment. Detecting deficiencies early allows for timely intervention and treatment.
Other tests performed alongside include complete blood count (CBC), methylmalonic acid (MMA), homocysteine levels, and Red Blood Cell (RBC) Folate Test.
Patient Preparation
The patient should avoid taking biotin supplements for at least 72 hours before sample collection. Fasting is required for 8 hours before the test.
Samples
1.5ml with a minimum volume of 0.6ml of serum collected in a transfer separated serum to a plastic transport tube if a red top tube is used. The sample should be refrigerated.

b12:180-1000pg/ml, folate >4.0ng/ml

Diseases / Diagnostic indicators
Some possible outcomes of the Vitamin B12 and Folate test include;
Low Vitamin B12 levels may indicate:
- Glossitis (inflamed tongue)

- Hyperreflexia (overactive reflexes)
- Ataxia (loss of coordination or balance)
- Megaloblastic anemia due to impaired DNA synthesis in red blood cells
Low Folate levels may indicate:

A normal result indicates sufficient levels of vitamin B12 and folate for healthy red blood cell function and nerve health.
Low levels may suggest dietary deficiencies, malabsorption, or underlying medical conditions affecting nutrient metabolism.
