Purpose
The progesterone test measures the amount of progesterone hormone in blood. Progesterone is a natural hormone made mainly by a woman’s ovaries after ovulation (when the egg is released each month).
It prepares the body for pregnancy by helping thicken the lining of the uterus so a fertilized egg can attach and grow. It also supports early pregnancy by keeping the uterus healthy for the baby. If there’s no pregnancy, progesterone levels drop, causing a period to start.
Men also have small amounts of progesterone that help with sperm production and balancing other hormones.
Patient Preparation
The patient should avoid taking biotin supplements for at least 72 hours before sample collection.
Samples
1ml with a minimum volume of 0.7ml of serum or plasma collected in a Gel-barrier, red-top, or green top tube (lithium heparin). Avoid using EDTA, oxalate, or citrate plasma. Transfer separated serum to a plastic transport tube if a red top tube is used. Store the sample at room temperature.

Male: 0.0-0.5 ng/ml, female non-pregnant:0.1-23.9ng/ml, pregnant:11.0-214.0ng/ml, menopausal:0.0-0.1ng/ml

Diseases / Diagnostic indicators
High levels are associated with congenital adrenal hyperplasia and decreased in hypogonadism and short luteal phase syndrome. Low levels are associated with infertility and menopause.
